چکیده

Phytosterols, their structure are very similar to that of cholesterol, have promise as a functional food material. However, they have not been actively used as material for low solubility in both oil and water. The research objective is to increase the solubility by modifying the sterol using microbial enzymes and to broaden the reach of phytosterols as biomass material and a food additive in our laboratory. We recently reported the characterization of novel cholesterol esterase from Trichoderma sp. AS59 isolated from soil. Therefore, in the present study, we examined the ability of the enzyme to synthesize steryl esters from sterol and free fatty acids of varying chain lengths. As a result, all the mediumand long-chain free fatty acids used are successfully employed in cholesterol ester synthesis and the yield of cholesteryl palmitate reached over 60% in large scale such as 30 mL of solvent. Of additional interest is that the fatty acid specificities in the synthesis and hydrolysis of cholesteryl esters were entirely different from each other. Furthermore, we attempted to synthesize stigmasteryl palmitate as a food additive and found that the enzyme catalyzed the synthesis of the ester. That indicates the potential utility of the enzyme in the food industry.

A simple method for the determination of the cholesterol esterase activity.

The proposed method determines the activity of cholesterol esterase (CEH) and takes advantage of its ability to catalyze the hydrolysis of cholesterol esters naturally present in human serum. The assay is based on Allain's method of spectrophotometric determination of cholesterol by means of cholesterol oxidase, peroxidase, but using 3,5-dichloro-dihydroxybenzenesulfonic acid (DHBS) as phenolic...

Fluorometric enzymatic determination of total cholesterol in serum.

We describe a fluorometric enzymatic method for determining total serum cholesterol, based on hydrolysis of cholesterol esters to free cholesterol by cholesterol ester hydrolase (EC 3.1.1.13). The free cholesterol formed, as well as that initially present, is then oxidized by cholesterol oxidase (EC 1.1.3.6) to cholest-4-en-3-one with simultaneous production of hydrogen peroxide. The latter cat...

Recombinant sterol esterase from Ophiostoma piceae: an improved biocatalyst expressed in Pichia pastoris

BACKGROUND The ascomycete Ophiostoma piceae produces a sterol esterase (OPE) with high affinity towards p-nitrophenol, glycerol and sterol esters. Its hydrolytic activity on natural mixtures of triglycerides and sterol esters has been proposed for pitch biocontrol in paper industry since these compounds produce important economic losses during paper pulp manufacture. RESULTS Recently, this en...

Enzymatic assay for cholesterol ester hydrolase activity.

A rapid and accurate method is described for the assay of cholesterol ester hydrolase (CEH) activity. Aliquots of the enzyme-substrate incubation mixture are extracted into isopropanol. The free cholesterol concentration in each extract is determined enzymatically using a single aqueous reagent containing cholesterol oxidase and peroxidase. The free cholesterol remaining after the cholesterol e...

Plant sterol and stanol substrate specificity of pancreatic cholesterol esterase.

Consumption of plant sterols or stanols (collectively referred to as phytosterols) and their esters results in decreased low-density lipoprotein cholesterol, which is associated with decreased atherosclerotic risk. The mechanisms by which phytosterols impart their effects, however, are incompletely characterized. The objective of the present study is to determine if pancreatic cholesterol ester...
